Genetics and Anti-Aging: NGS Panels for Predisposition to Age-Related Diseases
Genetic testing is playing an increasingly important role in anti-aging medicine. Polygenic risk scores (PRS) make it possible to assess genetic predisposition to cardiovascular disease, type 2 diabetes and Alzheimer's disease, while pharmacogenomics allows anti-aging therapy to be personalized.
NGS provides a platform for comprehensive genetic analysis, from targeted panels of 50–100 genes to whole-genome sequencing (WGS). For an anti-aging laboratory, this means being able to offer clients a genetic health passport.
Key Genetic Targets in Anti-Aging
Genes associated with longevity and age-related diseases:
- APOE: the key gene for assessing Alzheimer's disease risk. The ε4 allele increases the risk 3- to 15-fold.
- FOXO3: a regulator of autophagy and stress resistance. Its variants are associated with longevity.
- TERT: the telomerase gene. Polymorphisms affect telomere length.
- MTOR, SIRT1, AMPK: key regulators of metabolism and aging.
- CYP450 (pharmacogenomics): CYP2D6, CYP2C19, CYP3A4 for personalizing drug therapy.
- MTHFR, COMT, SOD2: genes involved in folate and catecholamine metabolism and antioxidant defense.
Equipment for Genetic Testing
The equipment set depends on the scale of testing:
- NGS sequencer: Illumina MiSeq Dx (clinical panels), NextSeq 2000 (extended testing, WES), Ion Torrent Genexus (fast turnaround).
- Automated DNA extraction: QIAGEN QIAsymphony, Promega Maxwell, Kingfisher Flex.
- Thermal cyclers: at least 2–3 units for amplification and library preparation.
- Fluorometer (Qubit) and bioanalyzer (TapeStation/Bioanalyzer) for DNA and library quality control.
- Bioinformatics pipeline: a local server or cloud solution for variant annotation and PRS calculation.
Integrating Genetic Data with Omics Approaches
Genetic testing delivers maximum value when combined with other omics data. For example, the APOE ε4 variant combined with elevated p-tau217 and an accelerated epigenetic age measured with epigenetic clocks forms a high neurodegenerative risk profile that calls for active prevention.
