Digital PCR
Digital PCR (dPCR) systems provide absolute quantification of nucleic acids without the need for calibration curves. The sample is partitioned into thousands to millions of micro-reactions, each undergoing independent amplification. Analysis based on the Poisson distribution yields the absolute copy number of the target. Key applications include oncology (liquid biopsy, monitoring of ctDNA and minimal residual disease), prenatal diagnostics (NIPT validation), virology (low viral load), gene therapy (AAV titration, CAR-T analysis) and transplantation medicine (rejection monitoring via donor-derived cfDNA). Compared with qPCR, digital PCR offers 10–100× higher sensitivity, full tolerance to PCR inhibitors and absolute accuracy without calibration curves.
